I want to make my inbox listing transparent like it shows in the preview for the new look but am unable to find this setting anywhere. Can anyone shed some light?

It isn't a setting - it's default behaviour for most views to be slightly transparent. 

You may have trouble with Internet Explorer though - there seem to be some elements of the new interface it can't display properly. 

Why not try Chrome or Firefox?

I tried several different themes, including the "Turf" one used in the preview. At the moment I'm using the "Wood" one as the contrast on the white inbox listings is in no way easy on the eyes. Which is a shame, I quite liked the "Pebbles" theme.

Same thing for me!  I want to make my inbox semi-transparent like in the preview to see the picture behind it, but I don't see that setting anywhere.

That's because it isn't a setting. Slight transparency is default behaviour.  

If your inbox is completely opaque than there is probably an issue with your browser. 

Try some basic troubleshooting things:

Clear cache and cookies
Try disabling any browser extensions and starting the browser again
Try starting GMail without any labs you might have enabled (use https://mail.google.com/mail/?labs=0 to do that)
Try another browser to see if the behaviour persists

Ah, yeah.  Tried in Firefox and the semi-tranparency works.  That's pretty bizarre that it isn't configured in a way the Internet Explorer can display it the same way.

I know partial transparency is the default behavior of most themes but is there any way that this can be customised so that the level can be increased?

Been trying to figure this out too. Just noticed on the bottom right side of the gmail page that there is a little link asking if you want to try the new look. I didn't see it at first because I had my gmail task list open. If there is something like gmail task list fixed to the bottom right side of your gmail page try getting rid of it. It might be covering up the option to try the new gmail.

Note: I also set my chrome browser theme to default and did the suggested steps above: 
Cleared the cache and cookies
Disabled all extensions and labs.
Then I restarted chrome.
